Genotoxicity of plant mediated synthesis of copper nanoparticles evaluated using in vitro mammalian cell micronucleus test

Objective: Nanotechnology is an emerging technology which has wide applications in many fields. Prime concern of research nanotechnology the synthesis nano-material with controlled size and shape. Recently, biosynthesis metallic nano-particles gained popularity owing to its eco-friendliness. The increasing use Copper nanoparticles (CuNPs) medicine industry demands understanding their potential toxicities. Methods: Genotoxicity was assessed using vitro micronucleus assay standard genotoxicity assay. In this study, nanoparticle tested absence presence metabolic activation (2% v/v S9 mix). Human peripheral blood lymphocyte cultures were exposed nanoparticle, at 5 dose-levels between 0.125 2 µL/mL culture medium system. Results: Required level cytotoxicity (55 ± 5% reduction replicative index, i.e., cytostasis) observed test concentration µL/mL. Therefore dose levels selected for scoring binucleated cells containing micronuclei were: 0.5, 1, From obtained data MNBN (Binucleated micronuclei) all three found range negative vehicle control. Conclusion: Our results concluded that, did not induce statistically significant or biologically relevant increase number activation.
